The transcriptional activator of the mannose phosphotransferase system (ManR) is a member of the BglG family of transcriptional activators which can exhibit complex regulation based on phosphorylation status. Extrapolating from the characterized MtlR system in Bacillus stenothermophilus, ManR can be phosphorylated at HIS-196 or HIS-266 by ManP resulting in decreased DNA binding and reduced mannose operon activation. When mannose is present, ManP preferentially phosphorylates mannose and dephosphorylates ManR resulting in transcriptional activation. ManR activation can be further enhanced by HPr-mediated phosphorylation of HIS-311 and HIS-370 resulting in increased DNA binding. This ability to calibrate transcriptional activator function based on the availability of substrates is a hallmark of the Escherichia coli antiterminator BglG with PTS regulation domains (PRD-I and PRD-II) serving analogous roles to the HIS-196/266 and HIS-311/370 regulation domains in ManR. Thus, BglG-family transcriptional activators are often misannotated as antiterminators due to their shared mechanism of regulatory control.
