Several transfer RNAs require modifications to provide maximal fidelity during protein synthesis. TrmR is an O-methyltransferase specific for 5-hydroxyuridines at tRNA position 34 generating 5-methoxyuridine ("mo5U" or "5"). TrmR is the second step of a two-step pathway resulting in sequential hydroxylation (TrhP1-TrhP2) and methoxylation (TrmR) steps targeting uridine(34). Staphylococcal tRNAs requiring this modification include: tRNAGUCC, tRNAVUAC, tRNAPUGG, tRNAAUGC, tRNAL3UAG, tRNATUGU, tRNASUGA.
