During conditions of oxidative stress, staphylococci protect free thiols in proteins by forming disulfides with bacillithiol, conceptually equivalent to glutathione. During recovery, bacilliredoxins like BrxA reduce protein-bacillithiol disulfides that are in the S-enantiomeric configuration back to free thiols. Thioredoxins (and oxidized bacillithiol) are then regenerated to an active (reduced) form by bacillithiol disulfide reductase (bdr/YpdA). Bacilliredoxins often display specificity in the target proteins that they reduce; BrxA is very efficient at reducing bacillithiolated glyceraldehyde 3-phosphate dehydrogenase (GapDH).
