Conversion of pyruvate to oxaloacetate is an essential step in multiple pathways from the citric acid cycle to gluconeogenesis and lipid synthesis. Staphylococcal pyruvate carboxylase (PC) is a homotetramer with each subunit containing five domains: a biotin carboxyl carrier protein (BCCP) domain which binds to the essential cofactor, a biotin carboxylase (BC) domain which modifies the BCCP-bound biotin to carboxybiotin and a carboxyltransferase (CT) domain which both stabilizes pyruvate as an enolate and releases CO2 locally from carboxybiotin, favoring production of oxaloacetate. The remaining two domains include an allosteric domain (AD) for regulation of catalytic activity and a PC tetramerization (PT) domain to facilitate the macromolecular assembly of the functional homotetramer.
