Ribosomal peptide release has not been extensively characterized in firmicutes and functions are therefore extrapolated from foundational studies in E. coli and pseudomonas. Peptide chain release factor 3 (RF-3), a "type 2" release factor, is structurally and functionally distinct from "type 1" release factors such as RF-1 and RF-2. Whereas Type 1 RFs promote release of completed peptides from the ribosome at stop codons, RF-3 serves a more regulatory and factor-recycling role. RF-3 stimulates dissociation of type 1 RFs from the ribosome after they have finished releasing the completed peptide. This is particularly important at UGA stop codons which effectively sequester RF-2 until it is released by RF-3. RF-3 may also enhance recruitment of type 1 RFs to the ribosome, further increasing the catalytic efficiency of peptide release and ribosome subunit recycling. Finally, RF-3 binds to, and is inhibited by, GTP, suggesting a regulatory role for the phosphorylation state of intracellular guanosine nucleotide pools in the rate of peptide product release and ribosomal recycling.
