MurA and MurZ can each catalyze the first committed step of cell wall synthesis - while either single-mutant is tolerated, a double-mutant is lethal. However, there are distinctions. MurZ is more resistant to fosfomycin inhibition. MurZ also appears to play a currently-undefined role alongside the dephosphorylated ReoMY system in directing MurA for ClpCP-mediated proteolysis.
