hup (hSau) : DNA replication initiation and transcriptional regulator protein HU [1]
• A crystal structure is available : 8HD5
Histone-like nucleoid-associated proteins are poorly-characterized DNA-binding proteins with many proposed functions, including condensing DNA into a nucleoid, protecting DNA from degradation and providing structure to support the formation of transcription bubbles. Although typically non-essential individually due to functional redundancy, HU is an essential protein in both S. aureus and B. subtilis. Extrapolating from studies in Bacillus subtilis, HU likely participates with DnaA in initiation of chromosomal DNA replication but has additional, undescribed essential functions. Additional evidence suggests HU can also participate in transcription regulation by wrapping DNA in such a way that exposes some promoters and prevents display of others. Formal nomenclature for this protein has not yet been established; the orthologue from E. coli is protein HU, the orthologue from Bacillus subtilis added "Bs" to make HBsu. Whether Staphylococcus aureus goes with "HU" or "HSau" nomenclature is not yet certain. Of note, the HBsu-specific lysine N-acetyltransferase YfmK does not have a clear orthologue in staphylococci so it is not clear if staphylococcal HU is acetylated.
