Elongation factor G (alt: translocase) binds to L7/L12 of the 50S ribosome. Upon peptide bond formation, the 30S and 50S subunits rotate relative to each other and the tRNA ends no longer bind to the same "PTC center site" (e.g. tRNA in the A site on the 30S is now in the P site on the 50S, tRNA in the P site on the 30S is now in the E site on the 50S). These hybrid A/P and P/E positions stimulate GTP hydrolysis in elongation factor G which undergoes a conformational change, inserting a domain that physically displaces the hybrid tRNAs (and thus the mRNA) to the "PTC center site" it occupies on the 50S (i.e. it "pushes" tRNAs from the 30S P site to the 30S E site to match its 50S E site binding). Elongation factor G then dissociates allowing for another round of protein elongation to occur. Antibiotics such as thiostrepton and fusidic acid inhibit Elongation Factor G activity.
