The 50S ribosomal large subunit has several processing checkpoints to ensure proper assembly before advancing to a larger multiprotein complex. One of these checkpoints occurs after binding of subunit L27 which has an N-terminal extension that, while required for L27 incorporation, blocks further 50S ribosome assembly. Ribosome-processing cysteine protease (Prp) cleaves this N-terminal extension from immature 50S ribosomes, clearing the L27 checkpoint and allowing 50S macromolecular assembly to continue.
