rny (cvfA) : ribonuclease Y [1]

Ribonuclease Y is a bifunctional riboendonuclease and a 2',3'-cyclic RNA monophosphate 2'-phosphodiesterase. Like most other riboendonucleases, RNase Y converts a terminal 3'-OH to a 2',3'-cyclic nucleotide monophosphate at the new terminus after cleavage. Both forms (3'-OH and 2',3'-c-P) are recognized and degraded by polynucleotide phosphorylase. Ribonuclease Y can also perform an additional step, cleaving RNA transcripts from a 3'-OH to a new 2',3'-c-P, but then hydrolyzing the terminal 2',3'-cyclic nucleotide monophosphate into a 3'-phosphate which blocks PNP degradation. As such, RNase Y was first identified as Conserved Virulence Factor A (CvfA), a gene critical for virulence in the silkworm infection model. In this role, RNaseY stabilizes agr transcripts as terminal 3'-phosphates, sharply increasing production of agr-regulated virulence traits such as hemolysin production. By regulating its cyclic nucleotide phosphodiesterase activity, RNase Y can contribute to both mRNA-stabilizing maturation as well as mRNA degradasome functions.
