The second staphylococcal core genome immune evasion cluster (IEC-2) encodes the dual-function extracellular fibrinogen- and complement C3b-binding immunomodulatory protein Efb. This protein prevents complement C5 activation and adaptive immunity through its C-terminal domain by forming higher order oligomers with cell surface-associated C3b and either itself or Efb. Through its N-terminal domain, it also binds to fibrinogen and platelets, cross-linking C3d-opsonized bacteria to a platelet-fibrinogen "shield" that prevents phagocytosis and can form a biofilm-like matrix. Because it only binds to C3b/d, Efb only prevents C3 deposition onto the staphylococcal cell by the alternative activation pathway, allowing opsonization and C3d processing by the classical or lectin pathways. However, membrane-bound C3d complement maturation into activated C5 convertase is blocked by Efb (or Ecb binding). C3b/C3d binding also prevents host B-cell and T-cell adaptive immunity via blocking of immune cell CR2 receptor/staphylococcal-opsonized-C3d coactivation. Finally, Efb released into a host immune cell's cytoplasm by intracellular staphylococci alters the immune activation cascade, blunting the inflammatory immune responses. Efb contains a signal peptidase secretion signal and is cleaved between alanine-29 and serine-30 to generate mature Efb.
