Staphylococci can produce up to 15 different capsules based on variants of the core trisaccharide N-acetyl-D-mannosaminouronate (ManNAcA) - N-acetyl-L-fucosamine (L-FucNAc) - N-acetyl-D-fucosamine (D-FucNAc). CapP catalyzes the first of two steps in the biosynthesis of UDP-N-acetyl-D-mannosaminouronate (D-ManNAcA), an epimerization at C-2 of UDP-N-acetyl-D-glucosamine to form UDP-N-acetyl-D-mannosamine.
Historically, different capsule types were assigned their own nomenclature (i.e. Cap5P and Cap8P for the two most prevalent capsule types, CP5 and CP8). The CapP protein is 99% identical between Cap5P and Cap8P, provides the same function, and is therefore harmonized as CapP.
