Staphylococci can produce up to 15 different capsules based on variants of the core trisaccharide N-acetyl-D-mannosaminouronate (ManNAcA) - N-acetyl-L-fucosamine (L-FucNAc) - N-acetyl-D-fucosamine (D-FucNAc). CapO catalyzes the second of two steps in the biosynthesis of UDP-ManNAcA, oxidizing the C-6 hydroxyl group of UDP-N-acetyl-D-mannosamine.
Historically, different capsule types were assigned their own nomenclature (i.e. Cap5O and Cap8O for the two most prevalent capsule types, CP5 and CP8). The CapO protein is 98% identical between Cap5O and Cap8O, provides the same function, and is therefore harmonized as CapO.
