Staphylococci can produce up to 15 different capsules based on variants of the core trisaccharide N-acetyl-D-mannosaminouronate (ManNAcA) - N-acetyl-L-fucosamine (L-FucNAc) - N-acetyl-D-fucosamine (D-FucNAc). CapN catalyzes the second of two steps in the biosynthesis of UDP-N-acetyl-D-fucosamine (D-FucNAc), reducing the C-4 ketone group of UDP-N-acetyl-dideoxy-D-xylo-hex-4-ulose.
Historically, different capsule types were assigned their own nomenclature (i.e. Cap5N and Cap8N for the two most prevalent capsule types, CP5 and CP8). The CapN protein is 97% identical between Cap5O and Cap8O, provides the same function, and is therefore harmonized as CapN.
