Staphylococci can produce up to 15 different capsules based on variants of the core trisaccharide N-acetyl-D-mannosaminouronate (ManNAcA) - N-acetyl-L-fucosamine (L-FucNAc) - N-acetyl-D-fucosamine (D-FucNAc). CapF catalyzes the second of three steps in the biosynthesis of UDP-N-acetyl-L-fucosamine (L-FucNAcA), a ketoreduction and C-3 epimerization of UDP-N-acetyl-2,6-dideoxy-beta-L-arabin-4-osamine to form UDP-N-acetyl-2,6-dideoxy-beta-L-talosamine.
Historically, different capsule types were assigned their own nomenclature (i.e. Cap5F and Cap8F for the two most prevalent capsule types, CP5 and CP8). The CapF protein is 98% identical between Cap5F and Cap8F, provides the same function, and is therefore harmonized as CapF.
