Staphylococci can produce up to 15 different capsules based on variants of the core trisaccharide N-acetyl-D-mannosaminouronate (ManNAcA) - N-acetyl-L-fucosamine (L-FucNAc) - N-acetyl-D-fucosamine (D-FucNAc). CapE catalyzes the first of three steps in the biosynthesis of UDP-N-acetyl-L-fucosamine (L-FucNAcA), a C-4,6 dehydrogenation and a C-5 epimerization of UDP-N-acetylglucosamine to form UDP-N-acetyl-2,6-dideoxy-beta-L-arabin-4-osamine.
Note: several strains will be capsule-deficient due to a nonfunctional CapE D75Y mutation characteristic of USA300 and USA500 lineages. They should be annotated as "nonfunctional capsule polysaccharide precursor UDP-N-acetylglucosamine 4,6-dehydratase/5-epimerase".
Historically, different capsule types were assigned their own nomenclature (i.e. Cap5E and Cap8E for the two most prevalent capsule types, CP5 and CP8). The CapE protein is 99% identical between Cap5E and Cap8E, provides the same function, and is therefore harmonized as CapE.
