capD : capsule polysaccharide precursor UDP-N-acetylglucosamine 6-dehydratase CapD [1]
Staphylococci can produce up to 15 different capsules based on variants of the core trisaccharide N-acetyl-D-mannosaminouronate (ManNAcA) - N-acetyl-L-fucosamine (L-FucNAc) - N-acetyl-D-fucosamine (D-FucNAc). CapD catalyzes the first committed step in biosynthesis of capsular polysaccharide monomer UDP-N-acetyl-D-fucosamine via C-4,6 dehydrogenation of UDP-N-acetylglucosamine.
Note: several strains will be capsule-deficient due to a nonfunctional CapD frameshift mutation characteristic of USA300 and USA500 lineages. They should be annotated as "nonfunctional capsule polysaccharide precursor UDP-N-acetylglucosamine 6-dehydratase CapD pseudogene".
Historically, different capsule types were assigned their own nomenclature (i.e. Cap5D and Cap8D for the two most prevalent capsule types, CP5 and CP8). The CapD protein is 99% identical between Cap5D and Cap8D, provides the same function, and is therefore harmonized as CapD.
